The following font is missing the @ sign. I would like to create a copy of the font file with my own @ symbol added to it:

Go to Insert > Symbol. Pick a symbol, or choose More Symbols. Scroll up or down to find the symbol you want to insert. Different font sets often have different symbols in them and the most commonly used symbols are in the Segoe UI Symbol font set.
To install the TrueType font in Windows:Click on Start, Select, Settings and click on Control Panel. Click on Fonts, click on File in the main tool bar and select Install New Font. Select the folder where the font is located. The fonts will appear; select the desired font that is titled TrueType and click on OK.
Fontforge should be able to do that. http://fontforge.github.io
